I was wondering if there is any other way that determines the appearance of the Subtitle in a DCP except the settings I make in DCP-O-MATIC?
Can the appearance (font/ size) be already determined in the srt ?
Or is the determinating factor the setup in DCP-O-MATIC?

No, I'm afraid not: SRT doesn't support font/size formatting but it does support formatting text for Italics by inserting the code <i> at the front of the text and closing with the code </i> at the end. You can apply the codes directly to the SRT text file or with your subtitling software. I think that you can also do bold formatting but I'm not really sure.
The ASS format does in fact support font/size as well as outline/shadow and positioning, but for now DCP-o-matic doesn't support the ASS format.

It might be slightly more accurate to say that DoM does support ASS, but there are some bugs with that support. I'm working on it.
